1321 Low temperature and mTOR inhibition favor human epidermal stem cell maintenance

Owing to their location at the interface with external environment, stem cells of epidermis are particularly exposed environmental variations such as fluctuating temperature. However, little is known about how temperature affects epidermal cell metabolism and behavior. Here we demonstrate that cultured human (HESCs) respond a small drop in through thermos-sensitive TRP channels connected mTOR signaling. Exposure HESCs rapamycin or induces nuclear translocation an impact on gene expression. We also by single-cell analysis long-term inhibition mTORC1 favors maintenance stemness. Taken together our results can adapt changes (e.g, temperature) signaling, constant maintenance, finding paramount importance for regenerative medicine applications.
